I had this problem back in April. This is what I did to fix it--dunno if it will work this time.
Look for a folder named "LoE" in your Local folder:
C:\Users\<your user name>\AppData\Local\
(Another user has posted that the folder should be in LocalLow.)
Rename or delete it.
Restart LoE. It should re-create the folder tree and re-download the assets (mostly talent marks).

Try going into your Local folder and renaming the LoE folder so that the game needs to recreate it.
That did it for me
